羊驼皮肤结构基因(原)胶原的表达特点

摘  要:为了揭示羊驼皮肤胶原蛋白(Collagen)在皮肤结构发生中的分子机制,本研究通过构建羊驼皮肤cD-NA文库并进行大规模测序分析,结果表明:在羊驼皮肤内发现只有纤维类胶原表达,即typeⅠ,typeⅢ,type Vcollagen,其中typeⅠ表达最高,typeⅢ和typeⅤ表达低;然而,在羊驼皮肤内未发现各类collagen相对应的原胶原(procollagen),其成员procollagen typeⅠ,typeⅢ,typeⅣ,typeⅥ,typeⅦ,typeⅩⅧ在表达,且typeⅠ远远高于其他家族成员的表达,由此推断collagen和procollagen typeⅠ在羊驼皮肤结构发生中起主要作用,羊驼皮肤内的蛋白水解机制可能使procollagen产生不同类型的collagen。Collagen is a big protein family, which is expressed highly in vertebrates and whose members are synthesized as inactive procollagen precursors which produce collagen by proteolysized. Collagen family providing structural support and strength to the extracellular matrixes of bone, cartilage, skin, blood vessels and most other connective tissues, forms the protective layer of skin and is the one of the factors deciding the skin structure. In order to find the molecular mechanism of collagen involving in the alpaca skin formation, cDNA library was constructed and sequenced. In the profile gene expression of alpaca skin, only fibrila collagen that was type Ⅰ, type Ⅲ, typeⅤ collagen were expressed and the expression of type Ⅰ collagen was highest. However, procollagens were not expressed corresponding to collagen. The type Ⅰ, type Ⅲ, type Ⅳ, type Ⅵ, type Ⅶ, type ⅩⅦ procollagen were found to be expressed. Moreover, the expression of type Ⅰ procollage was more higher than other members. From the results, it was deduced that type Ⅰ (pro)collagen played the main role in the structure formation of alpaca skin, and that the proteolysization mechanism could make procollagen pruduce different collagen.
