摘 要:【目的】研究新疆紫草毛状根理化因子对其生长的影响。【方法】采用固体培养法研究培养基、接种量及温度对新疆紫草毛状根生长的影响。【结果】(1)毛状根在不同培养基中的相对生长率及增殖倍数均表现接种量0.1 g较大。BS_5固体培养基的相对生长率及增殖倍数达最高,分别为1 790%和18.90倍。(2)毛状根在B_5培养基中培养33 d时鲜重达峰值,毛状根的继代时间不宜超过33 d。(3)26℃条件下毛状根的干重达最高,是接种量的19.2倍。高温处理毛状根的时间越长其受抑制程度越大。【结论】培养基、温度及接种量对毛状根的生长均有影响。研究为采用固体培养法长期保存新疆紫草毛状根提供了理论依据。[Objective]The effects of different physical and chemical factors on Arnebia euchroma Royle Johnst hairy root growth were studied. [ Methods ] For studying the effect on culture medium, inoculum concentration and temperature of Arnebia euchroma Royle Johnst hairy roots growth in solid medium. [Results] With 0.1 g inoculum concentrations giving the highest content of relative growth rate and roliferafion times of hairy mot for various culture medium. It showed that the highest of relative growth rate and roliferation times is 1 790% andl8.9 times with B5 solid medium. Hairy root fresh weight is maximum after 33 days of culture and subculture can not be more than 33 days in B5 medium. It is 19.2 times inoculum concentrations and dry weight of hairy root is the highest in 26℃ treatment. The results show that the longer the hairy mot was treated with high temperature, the greater it was inhibited. [ Conclusion] Hairy mot growth were affected with culture medium, inoculum concentration and temperature. The study provides a theoretical basis for preserving Arnebia euchroma Royle Johnst hairy roots for a long period by solid culture medium.
