Effect of Exercise Training and Puerarin on Bone Mineral Density,Cytokine,Bone Metabolism Index in Ovariectomized Rats

摘  要:目的探讨有氧运动配合葛根素对去卵巢大鼠骨密度、血清细胞因子和骨代谢相关指标的影响。方法选取3月龄雌性大鼠36只,去卵巢手术后随机分组并分别进行自然饲养、单一补充葛根素和有氧运动配合补充葛根素的实验;12周后处死,取左心室血浆并离心出血清,分别采用放射免疫法、酶联免疫法、生化分析仪测量各组血清细胞因子和骨代谢指标的含量;应用双能X线骨密度仪测量大鼠腰椎第2-4骨,胫骨和股骨的骨密度。结果有氧运动配合葛根素组大鼠腰椎第2-4骨、股骨和胫骨骨密度较安静对照组呈显著性提高(P<0.05);与安静对照组相比,单一补充葛根素组血清细胞因子指标TNF-а具有显著性差异(P<0.05),有氧运动配合葛根素组指标TNF-а、TGF-β呈非常显著性降低(P<0.01),指标IL-6显著性降低(P<0.05),血清骨代谢指标钙、磷和碱性磷酸酶均较安静对照组具有显著差异性(P<0.05)。结论有氧运动配合补充葛根素可有效增加去卵巢大鼠骨密度,降低血清细胞因子和骨代谢相关指标的水平,对绝经后骨质疏松症状的抑制具有积极影响。Objective is to study the effect of exercise training and puerarin on bone mineral density, cytokine, bone metabolism index in ovariectomized rats. The 3-roonth-old femal rats of thirty-six seleced were randomly diveded into control group, puerarin group and aerobic exercise with the puerarin group after ovariectomized, after 12 weeks, the rats were death by left ventricle and centrifugation the serum of plasma, using the Radioimmunoassay, Enzyme-linked immunosorbent assay and Biochemical analyzer measure the serum cytokines and Indicators of bone metabolism;using the Dual-energy X-ray absorptiometry measure the first 2-4 lumbar bone , tibia and femur. Result the first 2-4 lumber bone, tibia and femur in rats of aerobic exercise with Puerarin were significantly increased than quiet control group( P 〈 0.05) ; the TNF-a of serum cytokines in puerarin group was quiet singnifieant different than the control group( P 〈 0.05), the TNF-a and TGF-β in aerobic exercise were seemed very quiet significant different than control group (P 〈 0.01 ), and the IL-6 seemed significant different(P 〈 0.05); the serum calcium, phosphorus and alkaline phospbmase of bone metabolism in aerobic exercise with Puerarin group wereshowed significant differences than the quiet control group ( P 〈 0.05). Conclusion is that the aerobic exercise with Puerarin can effectively increase bone mineral density, reduced the serum cytokines and bone metabolism , which has a significant impact the inhibition of postmenopausal osteoporosis.
