摘  要:利用组织病理学研究方法对油茶炭疽病菌的侵入过程进行了观察。用整叶透明染色法观察到病菌分生孢子可在细胞的一端、两端发芽,芽管顶端逐渐膨大形成附着胞;有些芽管还可形成分枝,并在各分枝端形成附着胞,少数褐色的附着胞陷入叶背的气孔内。经石蜡切片观察到表皮细胞内的菌丝逐渐扩展到叶肉细胞内,形成大量菌丝体,并在寄主叶表皮下形成分生孢子盘,分生孢子盘成熟后突破叶表角质层外露。透射电镜观察到油茶叶片感染炭疽病后,其细胞结构发生变化,不同发病程度的组织中细胞的超显微结构有不同的反应。随着发病程度的加重,细胞器结构被破坏严重,叶绿体数量越来越少。有些发病严重的细胞还发生质壁分离现象,并可看到少量菌丝。Histopathological observation on invasion of Colletotrichum gloeosporioides to oil tea was conducted with using tissue clearing and staining, paraffin sectioning and transmission electron microscope. Through whole leaf tissue clearing and staining technique, it was clearly observed that the pathogen germinated and formed germ tubes from one or both ends of the cells and the tips of germ tubes gradually swelled to form appresoria;some germ tubes branched and formed appresoria at the tips of all branches and a few brown appresoria grew into the stoma on the back of leaves. Through paraffin sections ,it was observed that the mycelia in epidermal cells gradually extended in to mesophyll cells. Acer- vuli formed under the epidermis of the host and broke the cuticle layer of epidermis when it matured. It was also observed with transmission electron microscope that the cell structure of leaves changed due to the infection of pathogen. The structures of cells at different disease severity showed different reactions. As the disease level increased, the cell structure was severely damaged, and the number of chloroplast declined. Some severely damaged cells became plasmolysis, and mycelia within the cell could be observed.
