摘 要:目的:建立升麻中总三萜的含量测定方法,比较不同提取方法的提取效果。方法:采用紫外显色法,以0.20mL 5%香草醛-冰醋酸作为显色剂,0.6mL高氯酸作为稳定剂,70℃水浴反应15min,在543nm测定吸光度。结果:三萜皂苷含量在95.00μg^221.67μg内线性良好,r=0.9997,平均加样回收率为99.4%,RSD=0.698%(n=3)。分别以不同浓度的乙醇溶液提取升麻三萜,确定了最佳提取剂为75%乙醇。结论:该方法准确、简便、重现性好,可作为升麻中总三萜的含量测定方法。Objective: A method for the determination of total triterpenoids in Cimicifugae Rhizoma was established, extraction process with different concentrations of ethanol-water solvents was also optimized. Methods: A UV and colormetric analysis meth od was used applied with using 0. 20 mL 5% vanillin-vcetic acid glacial as developer, 0. 60mL perchloric acid as stabilizer under 543 nm in water bath at 70℃ for 15min. Results:The linear relationship of triterpenoids was good in a range of 95.00 g - 221.67 g (r=0. 9997) and average recovery was 99. 4%, RSD=0. 698% (n=3). Triterpenoids were extracted in different concentrations of ethanol water, and 70% ethanol was considered to be the best solvent. Conclusion:The method is convenient, accurate, reproducible and can be used for quality control of triterpenoids in Cimicifugae Rhizoma.
