大黄虫丸抗动脉血栓形成的基因表达谱研究

摘　　要：目的:探讨大黄虫丸对血小板基因表达谱的影响,从分子水平揭示大黄虫丸抗血栓的机理。方法:动脉血栓形成患者服用大黄虫丸一定疗程后,分离单核细胞与血小板,提取总RNA,利用含14 000个基因的基因芯片,分析大黄虫丸对动脉血栓形成患者血小板基因表达的影响。结果:服用大黄虫丸患者血小板有44个基因表达上调,299个基因表达下调;单核细胞有252个基因表达上调,466个基因表达下调,基因聚类分析发现这些基因涉及离子通道和运输蛋白、细胞凋亡相关的蛋白、DNA合成和修复、转录及转录因子、细胞受体、细胞信号和传递蛋白、蛋白翻译合成等方面。结论:大黄虫丸对动脉血栓形成者血小板与单核细胞基因表达谱有一定的调节作用,这些基因涉及离子通道和运输蛋白、细胞凋亡相关的蛋白、DNA合成和修复、转录和转录因子、细胞受体、细胞信号和传递蛋白、蛋白翻译合成等。Objective：To investigate the effect of Dahuangzhechong pill on the gene expression spectra of preventing arterial thrombosis,and reveal its mechanism on molecule level. Methods：Mononuelear cell and blood platelet of the arterial thrombosis patients were separated before and after treatment by Dahuangzhechong pill. Their RNA was extracted respectively and the genes expressions were detected using gene array containing 14 000 gene. Results：44 genes up-expressed and 299 genes down-expressed in blood platelet,252 genes expression increased and 299 genes expression decreased in mononuclear cell genes after treated with Dahuangzhechong pill. The cluster analysis showed that the genes contained ion channel and transport protein, apoptosis related protein, DNA synthesis, repair and transcriptio factor, cell receptor, cell signal and transducin, and protein translation and synthesis, etc. Conclusion：Dahuangzhechong pill may prevent arterial thrombosis through genes containing ion channel and transport protein, apoptosis related protein, DNA synthesis, repair and transcriptio factor, cell receptor, cell signal and trausducin, and protein translation and synthesis, etc.
