摘  要:目的预测肠道病毒71型(EV71)外壳蛋白VP1二级结构和B细胞表位,为制备EV71疫苗的研究提供理论基础。方法应用GOR4、HNN、SOPMA、nnPredict等方法预测EV71 VP1二级结构,并结合亲水性、柔韧性、表面可能性和吴氏抗原指数法综合预测潜在的B细胞表位。结果EV71 VP1二级结构以无规卷曲和β-片层为主,含有少量的α-螺旋,少见β-转角;EV71 VP1潜在的B细胞表位很可能位于第67-71位氨基酸残基。结论联合多种生物信息学方法成功预测了EV71 VP1二级结构和B细胞表位;此为制备EV71疫苗的研究提供了理论基础。Objective To predict the secondary structure and B cell epitopes of enterovirus type 71 viral protein I , so as to provide theory evidence for the study of vaccine preparation for EV 71. Methods The secondary structure of enterovirus type 71 viral protein I was analyzed using the GOR4,HNN,SOPMA ,and the B cell epitope of EV 71 viral protein I was predicted by combining the hydrophilicity, flexility, surface probability and Wu's antigenic index method. Resuits The secondary structure of EV 71 viral protein I showed random coils and β-sheet mainly,but a little helix and no β-turn. The B cell epitopes was probably located at the regions of 67-71. Conclusion The secondary structure and B cell epitopes of EV 71 viral protein I can be predicted by multi-methods, which provides theory evidence for the study of vaccine preparation for EV 71.
