摘  要:蛋白激酶C受体蛋白(RACK)除作为细胞内蛋白激酶C的受体之外,还作为细胞内重要的支架蛋白调节细胞功能。应用生物信息学方法对家蚕RACK蛋白的编码基因Bmrack进行电子克隆,获得了1224 bp的重组序列,基因编码区长960 bp,编码319个氨基酸,蛋白分子质量为36.04 kD,等电点为8.07。从家蚕幼虫脂肪体基因组DNA中PCR克隆了Bmrack基因,序列总长为2 122 bp,含有4个内含子和5个外显子,通过家蚕WGS比对,结果与其中5条序列的一致性为99%,且修复了其中8个缺口。聚类分析表明RACK蛋白的氨基酸序列在鳞翅目昆虫中非常保守。Apart from acting as receptor for protein kinase C C (RACK) is also an important scaffold protein to have other n cells,the receptor for activated protein kinase regulatory functions in cells. In present study, Bombyx mori rack gene (Bmrack) was in silico cloned based on bioinformatics and a contig of 1 224 bp was obtained. The length of its coding region was 960 bp and coded for 319 amino acids. Its molecular weight was 36.04 kD and pl was 8.07. After that, Bmrack gene was cloned from larval fat body genomic DNA through PCR and a sequence with 2 122 bp was obtained which contained 4 introns and 5 exons. Blasting against silk-worm whole-shotgun sequence (WGS) database yielded 5 genomic sequences of which sequence identities with Bmrack gene reached 99%. Multiple sequence alignment with these 5 sequences enabled us to identify nucleotides for 8 gaps existing in the WGS database. In addition, phylogenetic analysis showed that the amino acid sequences of RACK were highly conserved in Lepidoptera.
