摘 要:目的探讨益气温阳活血化瘀方药——心复康口服液对腹主动脉部分缩窄致压力超负荷模型大鼠心脏功能和心肌能量代谢的影响。方法将120只成年雄性SD大鼠采用随机数字表法分为3组:假手术组、腹主动脉缩窄模型组(模型组)、心复康治疗组(治疗组)。采用腹主动脉部分缩窄法制作压力超负荷大鼠模型,BL-420E+生物信号采集系统测定心功能参数,用高效液相法(HPLC)测定心肌高能磷酸盐的浓度,通过透射电镜观察大鼠心肌线粒体的形态,比较3组大鼠第6、8周心功能的各项指标(SAP、DAP、LVEDP、LVMI、+dp/dtmax、-dp/dtmax)及心肌高能磷酸盐(ATP、ADP、AMP、TAN、EC)的含量。结果①与假手术组相比,造模后第6周起,模型组大鼠左心室质量指数(LVMI)、左心室舒张末压(LVEDP)显著升高,左心室内压最大变化速率(+dp/dtmax)显著下降(P<0.01);心肌ATP、ADP、AMP含量显著降低(P<0.01);造模后第8周,上述改变更显著(P<0.01,P<0.05)。②和模型组相比,治疗组大鼠心功能指标及ATP、ADP、AMP、TAN的含量均明显改善(P<0.01)。③治疗组大鼠心功能指标及ATP、ADP、AMP、TAN的含量在8周时明显高于6周(P<0.01,P<0.05)。④治疗组大鼠心肌线粒体的受损程度较模型组轻。结论心复康口服液能明显改善慢性压力超负荷大鼠心肌能量代谢和心脏功能,并对心肌线粒体起到保护作用。Objective To investigate the effect of Xinfukang oral liquid on the cardiac function and levels of energy-rich phosphates in pressure overload-induced left ventricular hypertrophy rats. Methods Totally 120 SD rats were randomly divided into 3 groups, sham operation group (SH) ( n = 40), coarctation of abdominal aorta model group (CAA) (n = 40), and model and Xinfukang oral liquid treatment group (XFK) (n = 40). Parameters of cardiac function, levels of energy-rich phosphates and morphological changes of the myocardial mitochondria were observed at the 6th and 8th week after the treatment. Results In CAA group at the 6th week after operation, the cardiac function parameters (LVMI and LVEDP) were increased and + dp/dt was decreased, while ATP and ADP were decreased and AMP was increased in comparison with SH group (P 〈 0.01 ). These changes were much obvious in 8 weeks (P 〈 0.01, P 〈 0.05 ). Compared with those of CAA group, the parameters of heart function and energy-rich phosphates ( ATP, ADP, AMP, TAN) in XFK group were improved significantly (P 〈 0.01 ) at the 6th and 8th week. In XFK group, the parameters of heart function and energy-rich phosphates were much better at 8th week than at 6th week (P 〈 0.01, P 〈 0.05 ). The morphological change of mitochondria was lesser in XFK group than in CAA group. Conclusion XFK oral liquid significantly improves the myocardial energy metabolism in pressure overload rats and protects the function of myocardial mitochondria.
