脐血间充质干细胞支持下脐血单个核细胞扩增的研究

摘　　要：目的探讨脐血源间充质十细胞联合外源性细胞因子对脐血单个核细胞体外扩增的支持作用。方法从脐血中分离、培养出间充质干细胞并检测其细胞表面抗原；以此间充质干细胞作为细胞滋养层。将脐血单个核细胞接种于无血清培养体系中培养18天，在第0、7、10、14及18天检测有核细胞总数、CD34＋、CD133＋细胞数、集落形成单位数和（G2＋M＋S）期细胞含量变化。结果①从脐血中分离、培养出的间充质干细胞能稳定表达CD29、CD105和CD44，但不表达CD34和CD133；②外源性细胞因子及脐血源间充质干细胞均支持脐血间充质干细胞扩增，但以细胞因子联合脐血源间充质干细胞组效果最好，在第10天上述检测指标达到峰值，分别为第0天的（6．91±1．91）、（7．75±1.24）、（6．49±1．33）、（15．62±1．29）和（28．26±6．58）倍，且维持造血至少18天。结论①从脐血中能成功分离、培养血间充质干细胞，并完成细胞表型的初步鉴定；②外源性细胞网子联合脐血源间充质干细胞可有效扩增脐血单个核细胞。Objective To explore the effects on umbilical cord blood mesenchymal stem cells （UCB MSCs） combined with exogenous cytokines in supporting the expansion of umbilical cord blood mononuclear cells（UCB-MNCs）. Methods Mesenchymal stem cells （MSCs） were isolated from umbilical cord blood, and then were detected the cellular surface antigens. The UCB MNCs were cultured in a serum-free system for 18 days. On day 0, 7, 10, 14 and 18, total mononuelear cells,CD34 cells,CD133 cells,cellular cycle and hematopoietic progenitor cells were detected, respectively. Results （1） Mesenchymal stem cells which isolated and cultured from umbilical cord blood were negative for CD34 and CD133, but positive for CD29, CD105 and CD44. （2）Both exogenous cytokines and UCB-MSCs could support the expansion of UCB-MNCs. The expansion combined with UCB MSCs and exogenous cytokines had better effect than another groups. The above parameters reached the peak values on day 10. The culture system could maintain at least 18 days. Conclusion （1） MSCs can be isolated and expand cultured from umbilical cord blood. The preliminary phenotype identification can be accomplished. （2）UCBMSCs combined with exogenous cytokines can efficiently support the expansion of UCB-MNCs.
