摘  要:目的应用DNA水平聚丙烯酰胺凝胶电泳(PAGE)检测重庆地区汉族人MDR1C3435T基因的多态性。方法PCR扩增重庆地区正常汉族人外周血MDR1C3435T基因,以MboⅠ进行限制性酶切,用DNA水平PAGE作图谱分析。结果200名重庆地区汉族人MDR1C3435T基因中,表型T/T的频率为0.15,T/C为0.49,C/C为0.36。MDR1C3435T基因T频率为0.395,基因C为0.605。结论重庆地区汉族人MDR1C3435T基因的分布与我国其他地区人群基本接近,地区差异不明显。DNA水平PAGE为一种简单可靠的基因多态性分析方法,值得在基层单位推广。Objective To detect the MDRIC3435T gene polymorphism in Hans in Chongqing Region, China by DNA horizontal electrophoresis. Methods The MDRI C3435T gene was amplified from peripheral blood of healthy Hans by PCR and digested with Mbo I for mapping by DNA horizontal PAGE. Results The frequencies of genotypes T / T, T / C and C / C in 200 Hans were 0. 15, 0. 49 and 0.36, while those of genotypes T and C were 0. 395 and 0. 605, respectively. Conclusion The distribution of MDRIC3435T gene in Hans in Chongqing Region showed no significant difference with those in other regions in China. DNA horizontal electrophoresis was a simple and reliable method for analysis of gene polymorphism, which was worthy of popularization in grass-roots units.
