摘  要:为了检测半番鸭MC1R基因的多态性,并分析其多态性与半番鸭羽色性状的相关性,以羽色表型极端的黑白羽半番鸭为研究对象,首先采用克隆测序的方法寻找MC1R基因编码区的SNP突变位点,结果发现MC1R基因编码区存在7个SNP,其中同义突变A399G位点为限制性内酶切Hin61的识别位点。其次针对酶切位点,利用PCR-RFLP方法鉴定A399G位点的基因多态性,结果显示,白羽半番鸭存在AG和GG两种基因型,而黑羽半番鸭只表现出1种基因型AG。最后经卡方独立性检验分析,表明A399G位点的基因型与半番鸭羽色无显著相关(P>0.05),这可能与该突变位点未引起蛋白质相应部位的氨基酸发生改变有关。基因型GG是否为白羽半番鸭特有,尚需加大样本进一步分析验证。The study were to investigate the polymorphism of MC1R gene in white-feather and black-plumage mule duck populations,and analyze the relationship between polymorphism and feather color trait. Firstly,the single nucleotide polymorphism (SNP) was searched by means of DNA sequencing. There were 7 SNPs in MC1R gene exon,and the synonymy mutation A399G was identified by Hin61. Secondly,the polymorphism was detected using PCR-RFLP according to the SNP. The results showed that two genotypes AG and GG were observed in white-feather individuals, while only genotype AG in black feather individuals. Lastly,Chi-square test indicated that no significant association with the polymorphism of MC1R gene and feather color of mule duck was discovered and this result could be due to no change of amino acid in the SNP. The sample number shoud be increased to further validate that genotype BB was or not particular to the white mule duck.
