凝血酶介导肾小球系膜细胞细胞间粘附分子1表达上调及水蛭素的阻断作用

摘　　要：目的为了阐明凝血酶对肾小球系膜细胞细胞间粘附分子１（ＩＣＡＭ－１）表达的影响。方法利用体外培养的人肾小球系膜细胞，分别采用噻唑蓝掺入法、流式细胞术、间接免疫荧光、Ｎｏｒｔｈｅｒｎ杂交等方法分别观察了凝血酶对系膜细胞增殖和表达ＩＣＡＭ－１蛋白和ｍＲＮＡ的影响，并同时进行了人工重组水蛭素的阻断实验。结果凝血酶可以显著促进系膜细胞增殖以及系膜细胞ＩＣＡＭ－１蛋白和ｍＲＮＡ的表达，水蛭素可以特异地阻断凝血酶的这些作用。结论凝血酶不仅是肾小球内凝血过程的关键酶，而且具有促进系膜增殖和促进系膜细胞表达ＩＣＡＭ－１的作用。Objective In order to verify our hypothesis that thrombin could enhance the expression of intercellular adhesionmolecule1(ICAM1)inmesangial cells.Methods MTT incorporation,flow cytometry,indirect fluorescence,and Northern blotting hybridization were used to detect the effect of thrombin on the proliferation and ICAM1 expression of human glomerular mesangial cells cultured in vitro,as well as the inhibitory effect of recominant hirudin.Results Mesangial cell proliferation and the ICAM1 protein and gene expression of ICAM1 were both promoted by thrombin.Hirudin was effective to inhibit the above effects of thrombin on mesangial cells.Conclusion Thrombin is not only the key enzyme in the pathogenesis of intraglomerular coagulation but also might be a potent mitogen and a potent ICAM1 inducer of glomerular mesangial cells.
