环孢素A联合雷公藤单体T_4对人系膜细胞增殖及其表达、合成白细胞介素-6的影响

摘　　要：目的探讨环孢素Ａ（ＣｓＡ）和雷公藤单体Ｔ４（Ｔ４）的免疫抑制机制以及两药联合使用的效应，特别是小剂量ＣｓＡ、Ｔ４联合获得较强免疫抑制效应的可能性。方法用３Ｈ－ＴｄＲ掺入率法测定人系膜细胞（ＭｓＣ）增殖，用逆转录－聚合酶链反应（ＲＴ－ＰＣＲ）和ＥＬＩＳＡ法测定细胞ＩＬ－６ｍＲＮＡ表达及培养上清ＩＬ－６含量。结果ＣｓＡ和Ｔ４均剂量依赖性抑制系膜细胞增殖及ＩＬ－６合成，两药联合产生明显的协同作用，小剂量ＣｓＡ、Ｔ４联合与大剂量ＣｓＡ作用相当。Objective To investigate the mechanisms of immunosuppression of CsA and T4 and effects of these two drugs used combinedly in lower dose.Methods MsC proliferation was determined by 3 HTdR incorporation.IL6mRNA expression and protein synthesis were measured by RTPCR and ELISA respectively.Results Both CsA and T4 inhibited MsC proliferation and IL6 producion in a dosedependent manner.Combination of two drugs produced an additional effect.A lowerdose of CsA associated with T4 could reach the same degree of immunosuppression as a higher dose of CsA does.Conclusion Combination of CsA and T4 produces a synergistic immunosuppression.
