摘 要:为了研究神经节苷脂GM3(Ganglioside,GM3)对人早幼粒白血病细胞系HL60细胞诱导分化后中性糖脂的影响,将神经节苷脂GM3加到HL60细胞的DME/F12培养液中,观察其对HL60细胞的诱导分化作用及分化后细胞中性糖脂的变化。结果:细胞吞噬功能显著增强,非特异性酯酶活性升高,细胞形态也呈现与这种分化相一致的改变。神经节苷脂GM3诱导HL60细胞分化后细胞中性糖脂各组分的百分含量发生了显著改变,CDH、CMH及CQH显著增多,与分化前相比P<005;CXH显著降低,与分化前相比P<005。The purpose of the present study was to investigate the changes of the neutral glycosphingolipid composition of a human promyelocytic leukemia cell line HL 60 cells differentiated by gangliside GM 3, After HL 60 cells were cultured with exogenous GM 3 in DME/F12 medium,that the effects on differentiation and change of neutral glycosphingolipid composition of HL 60 cells were observed.Results:The percentage of phagocytosing and the non specific esterase(NSE) activity significantly increased.Cells showed morphologically that they were differentiated into monocyte macrophage cells.After HL 60 cells were differentiated by gangliside GM 3,the remarkable increase was CDH、CMH and CQH( P <0 05 compared with undifferentiated),CXH showed significant decrease( P <0 05 compared with undifferentiated).It is suggested that Gangliside GM 3 can change neutral glycosphingolipid composition of the HL 60 cells.
