摘 要:为建立测定血浆中连翘酯苷含量的高效液相色谱方法,将血浆样品经去蛋白处理,以乙腈-水-醋酸(18∶82∶0.41)为流动相,流速为1 mL/min,检测波长为332 nm,柱温为25℃,进样量为10μL,经Angilent ZORBAX SB-C18(5μm×4.6mm×250 mm)色谱柱分离,进行色谱分析。结果表明,本方法连翘酯苷的血浆浓度在0.05-200μg/mL内线性关系良好,精密度和重复性试验RSD均小于5%。本方法简便、灵敏、重现性高,可用于连翘酯苷的药动学和相对生物利用度的研究。To establish a method for the determination of forsythiaside in plasma, after dealing with the plasma protein sample,acetonitrtle-water-phosphoric acid (18 ; 82.5 : 0.41) was taken as the mobile phase at a flow rate of 1.0 mL/min. The detective wavelength was 332 nm. The temperature was 25℃ and the sample volume was 10μL. The Angilent ZORBAX SB- C18 column (5 μm × 4.6 mm ×250 mm) was taken as the column for the separation and then progresssd the chromatographic analysis. In this method the plasma concentration of forsythiaside in the glycoside 0. 005 to 200 tzg/mL was good linearity, precision and repeatability test RSD was less than five percent. The results showed that the method is simple, sensitive and highly reproductive, and it is suitable for the research of the pharmacokinetics and relative bioavailability of forsythiaside.
