摘  要:目的探讨脂联素(ADPN)对高糖环境下肾小球系膜细胞(MCs)增殖及细胞间粘附分子1(ICAM-1)表达的影响。方法将MCs分为三组:①正常组(5.5mmol/L葡萄糖),②高糖组(25mmol/L葡萄糖),③ADPN组(25mmol/L葡萄糖,2.5ug/mlADPN)运用MTT测定(24h,,后MCs的48h96h)增殖情况;再将MCs分为三组:①正常组(5.5mmol/L葡萄糖),②高糖组(25mmol/L葡萄糖),③ADPN组(25mmol/L葡萄糖2.5,,,,,57.5101520ug/mlADPN),作用48小时后运用ELISA法测定各组MCs上清液中ICAM-1的含量。结果高糖组抑制细胞增殖,加入ADPN后促进细胞增殖。ICAM-1在高糖组表达较高,加入低浓度ADPN对于ICAM-1表达影响不大,~20mg/LADPN组和高糖组相比,10ICAM-1的表达下降,差异有统计学差异(P<0.05),且呈剂量依赖性。结论低浓度ADPN可以促进MCs增殖;中等以上浓度ADPN可以抑制ICAM-1在MCs的表达。提示ADPN可能通过阻抑ICAM-1表达发挥对DN的保护作用。Objective To investigate the effect of adiponectin on proliferation and expression of ICAM-1 in rat mesanglal ceils (MCs) cultured with high glucose. Methods The MCs were allocated into 3 groups: normal glucose group (5.5 mmol/L glucose), high glucose group (25mmol/L glucose), ADPN group(25 mmol/L glucose, 2.5ug/ml ADPN), 24h .48h. 95h later, the prolifer ation activity of MCs was observed by MTT. The MCs were allocated into 3 groups: normal glucose group (5.5 mmol/L glucose), high glucse group (25 mmol/ L glucose), ADPN group(25 mmol/L glucose, 2.5, 5, 7.5, 10, 15, 20ug/ml ADPN), 48 hours later, the expression of ICAM-1 was measured by ELISA. Results Compared with NG group, MCs proliferation activity inhibited and the expression of ICAM-1 significantly increased in HG group. The proliferation of MCs was promoted in adiponectin groups. The expression of TGF-β1 was suppressed in adiponectin groups compared with HG group. Conclusion Low adiponectln will suppress the proliferation activity of MCs cultured in high glucose. The high glucose can promote the expression of ICAM-1.10-20 ug/ml adiponectin will decrease the expression of ICAM- 1. which implies adiponectin may protect the diabetic kidney by decreasing the expression of ICAM-1.
