摘 要:目的探讨氯化钴(CoCl2)对小鼠促肾上腺皮质激素(ACTH)垂体腺瘤细胞AtT-20中低氧诱导因子-1α(HIF-1α)、糖皮质激素受体(GR)和阿片黑素促皮质激素原(POMC)表达的影响及其意义。方法25、50、100、200μmol/LCoCl2处理AtT-20细胞诱导其发生低氧,MTT检测细胞增殖的变化,实时定量PCR和Westernblot分别检测HIF-1α、GR、POMCmRNA和蛋白的表达情况。结果与对照组比较,25、50、100μmol/LCoCl2处理AtT-20细胞24、48h均可促进细胞生长增殖,处理AtT-20细胞24h其HIF—hx、GR、POMCmRNA和蛋白表达水平增高,且呈浓度依赖性,差异有统计学意义伊〈0.05)。结论低氧可诱导ArT-20细胞中H1F-1α,GR和POMCmRNA和蛋白表达水平的增加。Objective To investigate the effect ofCoC12-induced hypoxia on the expressions of hypoxia-inducible factor-1α (HIF-1α), glucocorticoid receptor (OR) and opiomelanocortin (POMC) in ART-20 cells. Methods ART-20 ceils were exposed to different concentrations (25, 50, 100, and 200 μmol/L) of COCl2 to induce hypoxia. MTT assay was employed to assess the changes in the cell proliferation after the exposure, and real-time PCR and Western blotting were performed to detect the expressions of HIF-1α, GR and POMC genes at both the mRNA and protein levels. Results Compared with the cells in normoxic conditions, the AtT-20 cells exposed to COC12 treatment at 25, 50, and 100 μmol/L for 24 and 48 h showed obviously enhanced cell proliferation. COCl2 significantly increased the mRNA and protein expressions of HIF-1α, OR and POMC genes in a dose-dependent manner (P〈0.05). Conclusion CoCl2-induced hypoxia up-regulates the mRNA and protein expressions ofHIF-1α GR and POMC genes in AtT-20 cells.
