摘 要:【目的】不同形式的剪切力作用对维持细胞、组织及机体正常生理状态有重要意义,合适的加载装置是细胞力学研究的前提。参照传统平行平板流室的理论和方法,研究适用于体外细胞的流体剪切力加载装置。【方法】设计制作平行平板流室加载装置,通过多普勒超声测速技术对流室流速等流体特征进行检测;并通过对小鼠成骨细胞的加载实验,检测装置的可行性与可操作性。【结果】彩色多普勒检测结果显示该装置内流体流动较均匀,剪切力加载均匀准确,操作简单方便。【结论】本研究装置可用于体外研究成骨细胞等力学敏感细胞在形态学、生理生化等方面对剪切力的反应。[ Objective ] Shear force plays an important role in the normal physiological functions of cells, tissues and the whole organism, whereas suitable loading devices are the basis in the cell mechanics research. Therefore, the parallel plate flow chamber system was developed according to the classical theory and methods. [ Methods] The flow velocity characteristic was detected through color Doppler ultrasound, and the facility of the device was tested through applying fluid shear stress to osteoblast. [ Results ] The result of the Doppler's ultrasonic testing showed the fluid flows uniformly, and no turbulent flow was found from the inlet to the center of the flow chamber. Furthermore, the system was simple and convenient to use. [Conclusion] The parallel plate flow chamber can be used to study the morphology and physiological actions of mechanics sensitive cells in vitro, such as osteoblasts, vascular endothelial cells, airway smooth muscle cells and so on.
