不同剂量的氯胺酮对大鼠免疫功能和肿瘤转移的影响

摘  要:【目的】探讨不同剂量的氯胺酮对大鼠免疫功能和肿瘤转移的影响。【方法】Fischer344雄性大鼠100只随机分为5组:生理盐水组(Saline组);氯胺酮25mg/kg组(K25组);氯胺酮50mg/kg组(K50组);氯胺酮100mg/kg组(K100组);氯胺酮200mg/kg组(K200组),每组20只。予腹腔注射给药处理,24h后各组随机抽取10只大鼠进行检测淋巴细胞亚群CD3+、CD4+、CD8+、CD4+/CD8+、NK细胞(CD161a+)的数量及NK细胞活性,而余下大鼠予尾静脉注入MADB106肿瘤细胞,于3周后测大鼠的肺转移瘤的数量。【结果】与Saline组相比,K25组CD3+、CD4+、CD8+、CD4+/CD8+没有明显变化,而K200组均明显降低(P<0.01),K50、K100组淋巴细胞亚群CD3+、CD4+亦明显亦降低(P<0.05或P<0.01);与Saline组相比,所有氯胺酮组大鼠CD161a+的数量和NK活性明显下降(P<0.01),而肺肿瘤转移明显增多(P<0.01)。氯胺酮剂量与大鼠NK细胞活性的Pearson相关系数为-0.801,氯胺酮剂量与大鼠NK细胞数量的Pearson相关系数为-0.420,氯胺酮剂量与大鼠肺肿瘤转移瘤的Pearson相关系数为0.969。【结论】氯胺酮能够抑制机体的免疫功能,从而促进肿瘤转移的易感性,NK细胞活性及肺肿瘤转移发生率与氯胺酮剂量相关。[Objective] To investigate the effects of different doses of ketamine on the immune function and tumor metastasis in rats. [Methods] 100 Fischer344 male rats were randomly assigned into 5 groups: Normal saline group(group Saline), ketamine 25 mg/kg group(group K25), ketamine 50 mg/kg group(group K50), ketamine100 mg/kg group(group K100), ketamine 200 mg/kg group(group K200). 24 hours after intraperitoneal injection of ketamine or normal saline, 10 rats in each group were picked up randomly to detect of lymphocyte subsets CD3^+,CD4^+,CD8^+,CD4^+/CD8^+ and NK cell counts, NK cell activity, The other 10 rats in each group were intravenous injected MADB106 tumor cells. Tumor lung metastases were assessed three weeks later. [Results] Compare to group Saline, the numbers of CD3^+,CD4^+,CD8^+ lymphocytes and the ratios of CD4^+/CD8^+ showed no significant different in group K25, but decreased significantly in group K200 (P 〈 0.01),and the numbers of CD3^+,CD4^+ lymphocytes were reduced significantly in group K50 and K100 (P 〈 0.05 or P 〈 0.01). Compare to group Saline, the numbers of CD161a^+ and NK cell activity of rats decreased significantly in all groups dealt with ketamine(P 〈 0.01 ), while tumor metastases in lung were were dramatically increased (P 〈 0.01). The Pearson correlation coefficient between the doses of ketamine and NK cell activity, NK cell counts, tumor Lung metastases is -0.801 .-0.420 and 0.969 separately. [Conclusion] Ketamine can suppress the immune function of rat, thus contribution to the susceptibility of tumor metastasis. NK cell activity and tumor Lung metastases are closely related with ketamine doses.
