摘  要:目的:进行正常大鼠与气虚血瘀证大鼠血清的蛋白质组学初步研究,寻找气虚血瘀证的差异表达蛋白。方法:复合因素制作大鼠气虚血瘀证动物模型,利用二维凝胶电泳,软件分析及质谱鉴定等技术观察气虚血瘀证大鼠血清的蛋白质表达谱改变情况。结果:初步鉴定出气虚血瘀证大鼠血清与正常组相比升高的蛋白质是结合珠蛋白(haptoglobin)、补体C3(complement component C3)、ZN--α2-糖蛋白(ZN-alpha-2-glycoprotein)、免疫球蛋白λ轻链(immunoglobulin lambda light chain),降低的蛋白质是CD5抗原样蛋白(CD5 antigen-like)。结论:初步发现的差异蛋白可能与气虚血瘀证的形成、发生、发展有关。Objective:To explore the changes of rat serum proteins related with syndrome of Qi-deficiency and blood stasis and to clarify the molecular biomarkers of the syndrome. Methods: The rat models with Qi-deficiency and blood stasis were established by combined methods. The changes of serum protein expressing pattern were obeserved by two dimensional gel electrophoresis (2-DE), Image Master 2D Elite 6.01 analysis software and MALDI-TOF-MS. Results:By MALDI-TOF-MS and then with the search engine MASCOT, compared with the normal group, 6 proteins were identified, in which CD5 antigen-like was down-regulated, other 5 proteins including haptoglobin, complement component C3, etc. were up-regulated. Conclusion: The functions and features in these proteins had relevance to the pathological process of inflammation, immune regulation, which is consistent with the known occurrence and development of the syndrome of Qi-deficiency and blood stasis.
