清热祛湿益气方对肝损伤细胞凋亡相关蛋白表达的影响

摘　　要：目的观察清热祛湿益气方对刀豆蛋白A(ConA)诱导的急性肝损伤模型小鼠肝细胞凋亡相关蛋白表达的影响。方法Balb/c雄性小鼠50只,随机分为空白组,模型组,清热祛湿益气方低、中、高剂量组,每组10只,连续灌胃7天。采用Con A诱导复制小鼠急性肝损伤病理模型,观察不同剂量清热祛湿益气方(7.5g/kg、10.0g/kg、12.5g/kg)对肝细胞Bax、Bcl-2以及Caspase-3 mR-NA表达的影响。结果与模型组比较,清热祛湿益气方不同剂量可下调模型小鼠肝组织Bax、Caspase-3 mRNA的表达,并可上调Bcl-2的表达(P<0.05),随剂量增大,Bcl-2阳性率分别为17.56%、18.28%、26.36%。结论清热祛湿益气方可抑制肝细胞凋亡,对Con A诱导的急性肝损伤具有治疗作用。Objective To investigate the effects of Qingre Qushi Yiqi Formula （QQYF） on the expressions of the hepatocyte apoptosis proteins in acute hepatic injury of mice induced by concanavalin A （Con A）. Methods The pathologic model of acute hepatic injury induced by Con A was used for evaluating the therapeutic effects of QQYF in mice. The Balb/c mice were randomized into 5 groups： blank group,model group,the 7.5g/kg QQYF group, the 10.0g/kg QQYF group and the 12.5g/kg QQYF group. All preparations were administrated by intra-gastric infusion. After 7 days, the expression of the apoptosis proteins including bax,bcl-2 and caspase-3 mRNA were measured. Results Bax and Caspase-3 mRNA expression decreased and Bcl-2 expression increased （（P〈0.05） after treatment by QQYF. With the dose of QQYF increased, Bcl-2 positive rates were 17. 56,18.28,26.36. Conclusion QQYF could inhibit hepatocyte apoptosis ,and has a protective effect on acute hepatic injury in mice induced by Con A.
