RP-HPLC法同时测定五味首乌片中淫羊藿苷、大黄素和大黄素甲醚的含量

摘　　要：目的:建立RP-HPLC法同时测定五味首乌片中多指标成分的含量。方法:采用Akasil C18(4.6mm×250mm,5μm)色谱柱,以乙腈(A)-0.4%磷酸(B)为流动相,梯度洗脱(0min,30%A;15min,50%A;20～30min,75%A;35～40min,30%A),流速1.0mL·min-1,检测波长270nm,柱温30℃,进样量20μL;供试品溶液制备采用溶剂分级萃取。结果:淫羊藿苷、大黄素、大黄素甲醚线性范围分别为1.44～7.18,1.28～6.39,0.690～3.45μg;平均加样回收率(n=6)分别为101.6%,103.0%,99.55%。结论:本方法快速、灵敏,重现性好,适合于五味首乌片中淫羊藿苷、大黄素和大黄素甲醚含量测定。Objective：To establish an RP - HPLC method for simultaneous quantitative determination of icariin, emodin and physcion in Wuwei Shouwu tablets. Methods：The analytical column was Akasil C18 column（4. 6 mmx 250 mm ,5 μm）. The mobile phase was composed of acetonitrile（A） and 0. 4% phosphoric acid（B） with gradient elution（0min,30% A ; 15 min, 50% A ; 20 - 30 min, 75% A ; 35 - 40 min, 30% A） at a flow rate of 1.0 mL · min-1. The wavelength of UV detector was 270 nm and column temperature was 30 ℃. The solution with 20 μL was injected into the HPLC system for analysis every time. The sample solution was extracted with different solvents. Re- sults： The linear ranges of icariin,emodin and physcion were 1.44 -7.18,1.28 -6.39 ,and O. 690 -3.45 μg,re- spectively;Their average recoveries （ n = 6 ） were 101.6%, 103.0%, 99.55%, respectively. Conclusion： This method is sensitive, repeatable and suitable to determine the contents of icariin, emodin and physcion in Wuwei Shouwu tablets.
