摘 要:以巨尾桉优株萌芽条茎段芽器官为外植体的组培技术体系研究结果表明:芽器官不同采集季节接种污染率的排序为,冬<秋<夏<春,萌动率的排序为,冬<夏<秋<春季。控制其外植体褐变最有效的措施是在初始培养基内添加8 mg/LVC+15 mg/L VB2,接种前期暗培养10天。增殖培养基中添加6-BA0.4 mg/L+NAAmg/L的激素组合,接芽18颗/瓶,培养20天,其芽的增殖系数达3.5,且芽体健壮。为了降低成本,提高经济效益,在巨尾桉优株的组培苗产业化育苗中,宜选择继代周期20天,高约1.0 cm的单芽苗株进行生根培养,而最适用的激素组合是在其生根培养基中添加ABT62.0 mg/L+IB I 1.0 mg/L,生根率达96%以上。Using bud organ in sprout of plus tree as the explant,the tissue culture of Eucalyptus grandis×E.urophylla was studied.The results showed that the contamination rate of inoculation of bud organ collected in different seasons ranked as winter 〈 autumn 〈 summer 〈 spring,and the rank of germination rate was winter 〈summer〈autumn〈spring.An effective technique for preventing browning is adding 8 mg/LVC and 15 mg/L VB2 to initial medium,and conducting dark culture of 10 days before inoculation.Adding 6-BA 0.4 mg/L and NAA 0.3 mg/L to proliferation medium,inoculated 18 buds/bottle,culture for 20 days,the proliferation index was 3.5,and the bud is health and vigorous.In order to lower the cost and enhance economic returns,during the course of industrialized tissue culture of plus tree of Eucalyptus grandis×E.urophylla,the suggested cycle for successive transfer culture is 20 days,using the single bud of 1.0 cm,adding ABT6 2.0 mg/L and IBA 1.0 mg/L to rooting medium,the rooting ratio can be 96 % or above.
