摘　　要：目的了解特发性血小板减少性紫癜(ITP)外周血单个核细胞(PBMCs)Toll样受体(TLRs)的mRNA表达情况。方法采用RT-PCR方法(SYBR GreenⅠ荧光染料结合法),对30名ITP患者和30名健康者的PBMCs中所表达的TLR2、4、6、7、9 mRNA进行相对定量检测。结果ITP组TLR2和TLR7的mRNA相对表达量均明显高于对照组(P<0.05),而TLR4、TLR6、TLR9的mRNA相对表达量两组间无明显差异(P>0.05)。结论TLR2和TLR7在ITP患者中表达水平上调,可能在ITP发病机制中起重要作用。Objective To study the level of the Toll like-receptor mRNA expression on PBMCs from patients with the idiopathic thrombocytopenie purpura. Methods We performed quantitative real-time reverse transeription-polymerase chain reaction （SYBR Green I fluorescent dye combination technique）analyses of PBMCs from 30 idiopathic thromboeytopenie purpura （ITP） patients and 30 healthy subjects, to estimate the relative levels of TLR2, TLR4, TLR6, TLR7 and TLR.9 mRNA expression. Results Relative expression levels of TLR2 and TLR7 mRNAs in ITP group （ n = 30） were significantly higher than those in health control group（ n = 30） （P 〈 0.05 ） , and relative expression levels of TLR4,TLR6 and TLR9 mRNA showed no significant difference between the two groups（P 〉 0.05 ）. Conclusion Expressions of TLR2 and TLR7 mRNA are up-regulated in patients with ITP, which may play an important role in the pathogenesis of ITP.
