摘 要:目的:建立一种适用于膜片钳技术的大鼠海马锥体神经元急性分离方法,并对其离子通道进行描述。方法:采用酶加机械分离法制备10~12 d鼠龄的大鼠海马锥体神经元,用全细胞膜片钳技术测定其生理学特性。结果:分离出的神经元形态正常,有较长突起;用膜片钳技术证实,其保存了主要的离子通道活性。结论:建立一种简单快速的适用于膜片钳技术的大鼠海马锥体神经元急性分离方法。Objective:To develop a modified method of acute isolation of rat hippocampal pyramidal neu- ron, and observe its ion current with patch clamp. Methods: The hippocampal pyramidal neurons were obtained by a combination of mechanical and enzymatic means from 10 -- 12 day - old rats. Whole - cell patch clamp tech- nique was used to measure the electrophysiological properties of the neurons. Results: The neurons isolated in this way had regular configuration. The neuraxons of them were very long. With patch- clamp techniques, we found that the methods could save the activities of primary iro.n channels. Conclusion: A simple and fast method for acutely isolating rats hippocampal pyramidal neuron suitable for patch clamp is successfully found.
