丁香提取物结肠定位包衣片在家兔体内的药物动力学研究

摘　　要：目的:考察丁香提取物的结肠靶向制剂和普通片剂中丁香苦苷体内药动学的特征,以便为其靶向性提供科学依据。方法:给家兔吞服两种制剂后,于不同时间采取血样,用高效液相色谱仪测定血药浓度,通过DAS2.0药动学软件自动拟合数据获得药动学参数。结果:家兔给予两种制剂后丁香苦苷药动学行为符合二室模型,结肠靶向制剂血药浓度出现时间晚于普通制剂,滞留时间约为4.6h。结论:药物动力学实验数据表明,丁香提取物的结肠定位包衣片具有良好的结肠靶向性。Objective: To investigate the pharmacokinetics property of Syringopicrosid in the tablets of colon-specific delivery coated and naked in vivo so that to provide the scientific data for the target effect. Methods: After orally administrated to rabbits, the blood samples were taken during the different times and the Syringopicrosid concentration in blood samples were determined with HPLC instrument. And then the pharmacokinetics parameters were taken by auto-fitting data with DAS2.0 pharmacokinetics software. Results: The Syringopicrosid in two preparations shows to be 2-compartment model after orally administrated to rabbits. The time appeared drug concentration in rabbit plasma to colon-specific delivery preparation was later than common tablet and the Tlag was about 4.6. Conclusion: The pharmacokinetics parameters showed that the colon-specific delivery tablet has good target effect in colon.
