摘  要:目的:研究绞股蓝总皂甙(GPs)、曲匹地尔(trapidil)对兔髂动脉内皮损伤后内膜增殖的抑制作用,以及上述过程中sis基因表达的改变及药物的影响.方法:用Fogarty球囊导管制成兔髂动脉内皮损伤模型,分别给予(GPs,traPidil治疗,于不同时间点进行病理形态学检查、平滑肌细胞肌动蛋白(SMCsactin)免疫组化染色及sis基因原位杂交(ISHH).结果:术后10d,28d损伤血管内膜明显增生、增厚,管腔狭窄,血管中膜及增生内膜有大量actin染色阳性细胞;ISHH显示sis基因mRNA明显表达;GPs,tra-Pidil可明显抑制损伤血管内膜的增生[内膜面积分别为(0.095±0.035)mm2,(0.065±0.014)mm2vs(0.184±0.063)mm2,P<O.05],但对sis基因表达无明显影响.结论:GPs对在体兔具有明显的抑制实验性动脉内皮损伤后内膜增殖的作用.Aim :To determine if gypenosides (GPs) could inhibit SMCs proliferation and influence sis gene expressionafter experimental artery deendothelialization. Methods:Both GPs and trapidil were administered before and afterballoon deendothelialization in rabbit iliac artery. Mor-phometry,SMCs actin immunohistochemical staining and sisgene in situ hybridization histochemistry (ISHH) were per-formed on the sections obtained from arterial segments. Re-sults: The intimal hyperplasia and luminal stenosis could befound in injured arteries 10 d, 28 d after balloon denuda-tion. The areas of neo-intima in GPs group and trapidilgroup were significantly smaller than those in control(0. 095±0. 035)mm2 and (0. 065 ± 0. 014)mm2 μs (0. 184 ±0. 063) mm2, respectively, P< 0. 05 ). Numbers of SMCsactin-positive staining cells appeared in neo-intima and me-dia in 10 d, 28 d after arterial injury. Remarkable expression of sis gene in media and neo-intima of injured arterieswas detected by ISHH in 3 d, 10 d, and 28 d after balloondenudation with intimal hyperplasia. GPs and trapidil hadno effects on expression of sis gene. Conclusions: The results suggest that GPs have obvious inhibiting effects on in-timal hyperplasia in rabbit iliac artery after balloon arterialdenudation.
