摘 要:目的观察Fas抗体、白介素1β(IL-1β)对体外培养的气道上皮细胞凋亡的影响。方法分离培养兔的气道上皮细胞(AEC),加入IL-1β与不同浓度的Fas抗体共同孵育,以流式细胞术和TUNEL法检测AEC凋亡的变化。结果以流式细胞术和TUNEL法检测,Fas抗体对AEC凋亡呈剂量、时间-效应依赖性作用;IL-1β对Fas抗体诱导AEC的凋亡没有明显影响。结论Fas抗体能够有效的诱导正常和炎症状态下原代AEC发生凋亡,可能对抑制气道炎症和气道重塑有积极作用。Objective To investigate the effect of Fas antibody(Fas Ab) and interleukin-1β(IL-1β) on apoptosis of cultured rabbit airway epithelial cell(AEC) in vitro. Methods The cultured AEC in vitro from rabbit were incubated Fas Ab at various concentration with or without IL-1β. The apoptotic rate of AEC induced by Fas Ab with or without IL-1β was measured and analyzed by flow cytometry (Annexin-FITC and PI staining) and TUNEL. The AEC treated by Fas Ab for 12 h,24 h,36 h were cultured and measured by flow cytometry. Results The Fas Ab can induce apoptosis of AEC in dose-and time-dependant manner. There was no significance between with IL-1β group and without IL-1β group. Conclusions Fas Ab can effectively induce apoptosis of primary AEC in the condition of normal or inflammation. IL-1β can be little effect in the induction of AEC apoptosis.
