摘 要:目的探讨护津方对细菌脂多糖(LPS)诱导急性肺损伤大鼠肺组织核转录因子-κB m RNA(NF-κBm RNA)表达的影响。方法40只雄性W istar大鼠随机分为正常对照组、模型组、地塞米松组和护津方组,每组再分为4h和8h两个亚组;分别用地塞米松和护津方在LPS诱导急性肺损伤模型前预处理大鼠。采用免疫组化法和定量RT-PCR检测大鼠肺组织NF-κB蛋白及NF-κB m RNA的表达,光镜观察肺组织病理形态。结果与模型组相比,护津方组两个亚组大鼠肺组织NF-κB蛋白及NF-κB m RNA的表达均显著降低。病理学观察显示,模型组大鼠肺组织出现大片出血及坏死,护津方组大鼠肺组织病理损伤程度较模型组减轻。结论护津方对内毒素致急性肺损伤大鼠有保护作用,其机制可能与其降低内毒素致急性肺损伤大鼠NF-κB蛋白及NF-κB m RNA的表达有关。Objective: To investigate the effect of Hujin Decoction on expression of nuclear factor-κB (NF-κB) mRNA in rats with acute lung injury induced by lipopolysaccharide (LPS) . Methods: Forty male Wistar rats were randomly divided into 4 groups: normal control group, model group, dexamethasone group and Hujin Decoction group. Each group had 4h and 8h two subgroups. All groups, except normal control group, were administrated with LPS by intravenous injection to induce acute lung injury. The rats in dexamethasone group and Hujin Decoction group were treated by dexamethasone and Hujin Decoction respectively before LPS induced. The animals were killed after injection of LPS for 4h and 8h, and then the expression of NF-κB mRNA and protein were determined by RT-PCR and immunohistochemistry. The histopathology of the lung injury was observed by light microscope. Results: Compared with model group, the expression of NF-κB mRNA and protein in the lung tissue were obviously decreased in the two subgroups of Hujin Decoction group. Pathological observation indicates that pulmonary hemorrhage and necrosis were serious in the model group and a better condition in the Hujin Decoction group. Conclusion: Hujin Decoction has protective effects on acute lung injury induced by endotoxin in rats. The drug maybe take effect by means of inhibiting the expression of NF-κB mRNA and protein in lung tissues.
