摘  要:目的观察三七总皂苷对内毒素休克大鼠血清TNF-α的影响。方法将SD大鼠随机分为生理盐水组、内毒素组、三七总皂苷大、小剂量组,分别予生理盐水、生理盐水、三七总皂苷小剂量液、三七总皂苷大剂量液腹腔注射,于末次注射后1h时麻醉。以生理盐水对生理盐水组进行尾静脉注射,并以内毒素对其余三组进行尾静脉注射,于注射后30m in、90m in、2h、4h、6h各时间点每组随机取6只大鼠,心脏取血测血清TNF-α。结果在注射内毒素各时间点上,各组血清TNF-α水平逐渐升高,至2h达高峰,4h时回落,各组血清TNF-α水平明显高于生理盐水组,且内毒素组明显高于三七总皂苷大、小剂量组;至6h时,三七总皂苷大、小剂量组血清TNF-α水平已恢复正常,但内毒素组血清TNF-α水平仍明显高于其它三组。结论三七总皂苷可明显降低内毒素所致炎症反应。Objective: To investigate the effect of total saponins of Radix notoginseng (RNS) on serum TNF-α in endotoxic shock trats. Methods: 120 clean grade and healthy SD rats were randomly divided into normal saline (NS) group, endotoxin (LPS) group, RNS low and high dose group, 30 in each group. Respectively, the groups were given NS, NS, low and high dose RNS according to the above-mentioned order by intraperitoneal injection. At lh after last time injection, all rats were anaesthetized then the NS group was given NS, and the other 3 groups were given LPS, by tail vein injection. At different time points after injection 30min, 90rain, 2h, 4h and 6h, each group was randomly taken 6 rats for heart blood to determine serum TNF-α. Results: After endotoxin injection at 30min, 90min and 2h, the serum TNF-α gradually increased, and reached the peak at 2h. It began to decrease at 4h. The serum levels of TNF-α in NS group was lowest, and the LPS group was highest. At 6h, the serum levels of TNF-α in RNS low and high dose group TNF-α had returned to normal, but that of LPS group was still significantly higher than the other 3 groups. Conclusion: RNS could be significantly reduced endotoxin induced inflammatory response.
