摘  要:目的探讨中药复方清热颗粒剂对耐环丙沙星铜绿假单胞菌外排泵耐药机制的影响。方法建立急性耐环丙沙星铜绿假单胞菌腹腔感染兔动物模型,予复方清热颗粒药液灌胃,空白对照组则给予等量蒸馏水灌胃。末次给药后无菌心脏取血,离心分离出含药血清和空白血清。应用外排泵抑制剂(CCCP)选择出外排泵阳性的耐环丙沙星铜绿假单胞菌9株,应用琼脂对倍稀释法测出在有含药血清、空白血清、复方清热颗粒、泵抑制剂的情况下环丙沙星对上述9株铜绿假单胞菌的最低抑菌浓度(M IC)变化。结果中药复方清热颗粒含药血清可以降低环丙沙星对铜绿假单胞菌的M IC值。结论复方清热颗粒含药血清可抑制铜绿假单胞菌的外排泵,从而改善铜绿假单胞菌对环丙沙星的耐药。Objective: To observe the effect of compound clearing-heat granules on the CIP resistance mechanism of efflux pump in pseudomonas aeruginosa. Methods: The rabbits' models infected CIP resistance pseudomonas aeruginosa were given compound clearing-heat granules. The control group was administrated equivalent distilled water. After the last drug, the drug serum and the blank serum were separated from heart blood by centrifugation. Then, 9 CIP resistance pseudomonas aeruginosas with positive efflux pumps were chosen by CCCP. With drug serum, blank serum, compound clearing-heat granules and CCCP, the MIC of these 9 strains were detected by agar dilution method. Results: The MIC of pseudomonas aeruginosa was decreased by the serum with compound clearing-heat granules. Conclusion: The serum of compound clearing-heat granules could inhibit efflux pump in pseudomonas aeruginosa in order to improve the resistance of the pseudomonas aeruginosa with CIP.
