摘 要:对草原红牛m-calpain的30 ku调节亚基的DNA序列通过PCR扩增得到1 830 bp大小的产物,这个产物序列包括从外显子4到外显子7的全部序列(包括它们之间的内含子序列)。利用限制性内切酶HhaⅠ对PCR产物进行酶切,并利用最小二乘线性模型初步探索了不同基因型与肉质性状间的关系。结果表明:在31头草原红牛群体中检测到2个等位基因A和B,2个等位基因的频率分别为0.355和0.645;检测到3种基因型AA、BB和AB,3种基因型的频率分别为0.161,0.452和0.387。草原红牛m-calpain基因中BB基因型对牛肉嫩度剪切力值有较大的影响(P<0.05);AA基因型对大理石花纹评分值有较大的影响(P<0.05)。The 1 830 bp fragment encoding for the regulatory subunit (30 ku) of China red cattle m - calpain gene, putatively including the sequences of exon 4 through exon 7, was amplified by PCR. Two alleles( A and B) of the China red cattle cattle m - ealpain regulatory subunit gene were identified with PCR - RFLP analysis using endonuclease Hha Ⅰ , which the frequency of alleles found was 0. 355 for A and 0. 645 for B. Three genotypes AA, BB and AB were identified among the 31 China red cattle breeds. The frequency of genotypes found was 0. 161 for AA, 0. 452 for BB and 0. 387 for AB. The association between genotype markers and meat traits were analyzed by general linear model at the level of DNA. The result was the BB genotype of China red cattle m - ealpain gene showed a highly significant negative effect on shear force of beef tenderness traits ( P 〈 0.05 ). The result was the AA genotype of China red cattle m - ealpain gene showed a highly significant positive effect on marbling score of beef tenderness traits ( P 〈 0.05 ).
