摘 要:目的观察益智汤对APP695转基因小鼠行为学及脑组织β淀粉样蛋白(Aβ)及其前体蛋白(APP)的影响,探讨益智汤治疗阿尔茨海默病(AD)的机理。方法3月龄APP695转基因小鼠随机分为模型组、益智汤组,同月龄同背景转基因阴性小鼠为正常组。益智汤组给予益智汤0.1 g/kg灌胃,模型组和正常组给予等体积蒸馏水。益智汤治疗3个月后,使用电迷宫检测各组小鼠学习记忆能力,采用免疫组织化学方法观察各组小鼠大脑皮层、海马APP和Aβ的表达情况。结果电迷宫检测发现,模型组小鼠学习记忆能力低于正常组(P<0.05),益智汤组小鼠学习记忆能力较模型组增强(P<0.05)。免疫组织化学结果提示,模型组小鼠脑组织各区APP和Aβ的表达高于正常组(P<0.01),益智汤组小鼠脑组织各区APP和Aβ的表达比模型组明显减弱(P<0.05)。结论APP695转基因小鼠是研究AD的良好模型,益智汤通过下调APP和Aβ的表达来改善其学习记忆能力。Objective To observe the influences of Yizhi Decoction on the ethology and the expressions of brain β-amyloid (Aβ) and amyloid precursor protein (APP) in APP695 transgenie mice, and to discuss the mechanism of Yizhi Decoction for treating Alzheimer' s disease (AD). Methods APP695 transgenic mice (3 months old) were randomly divided into the model group and Yizhi Decoction group, and mice of the same age and background with negative transgene were included into the normal group. The mice in Yizhi Decoction group were given intragastrieally Yizhi Decoction (0. 1 g/kg), and the model group and normal group were given equal quantity of distilled water. After 3 months the learning and memory ability of mice were examined by using electric maze test, and the expressions of Aβ and APP in mouse cerebral cortex and hippocampus were observed by applying immunohistochemical assay in all groups. Results The outcome of electric maze test showed that the learning and memory ability of the model group was lower than that of the normal group (P 〈 0.05 ), while that of Yizhi Decoction group was improved (P 〈 0.05). The result of immunohistochemical assay indicated that the expressions of Aβ and APP in the model group were higher than those in the normal group (P 〈0.01 ). The expressions of Aβ and APP were reduced significantly in Yizhi Decoction group compared with those in the model group ( P 〈 0.05). Conclusion APP695 transgenic mice can be taken as a good model for studying AD. Yizhi Decoction can improve learning and memory ability through down-regulating the expressions of Aβ and APP.
