硫酸肼还原法测定血清中硝酸盐浓度

摘  要:目的建立一种简便可靠的血清硝酸盐浓度测定方法。方法在碱性介质中用硫酸肼将去蛋白血清中的硝酸盐还原为亚硝酸盐,再用Griess试剂测定亚硝酸盐的浓度。结果在NaOH介质(pH13.2)中,硫酸肼有最佳的还原率,56℃水浴10min,反应即可完成。该法批内CV3.58%(41.6±1.49μmol/L,n=15),批间CV6.40%(79.8±5.11μmoL/L,n=10);平均还原率94.7%;平均回收率95.1%;线性范围10-200μmol/L;参考范围32.7±15.0(x^-±s,μmol/L)。结论该法还原反应快速、高效,重复性好,且操作简便,试剂价格低廉。Objective To establish a simple and rapid method for detecting nitrate in serum. Methods In deprotein serum, nitrate was reduced to nitrite by bydrazine sulfate in alkaline medium,then the nitrite was detected with diazotization assay. Results The optimum assay condition was incubated at 56℃ for 10 minutes in 1.25 mmol/L NaOH medium. The linearity was up to 200 μmol/L. The inter-and intraassay CV were 3.58%(41.6±1.49μmol/L,n= 15) and 6.40%(79.8± 5.11 μmol/L,n= 10) respectively. The average reduction rate was 94.7% ,and the mean recovery was 95.1%. The reference value range was 32.7±15. 0(x^-±s,μmol/L,n=28). Conclusion This method is simple,rapid and low-cost. It provides with a higher reduction rate and satisfactory accuracy.
