摘  要:以成年大鼠脊髓片骶髓后连合核(DCN)为模型,应用盲膜片钳全细胞记录法,研究了DCN神经元对与伤害性信息传递和抑制有关的递质的反应。证明:谷氨酸在DCN神经元引起了由NMDA和非NMDA受体介导的内向电流;P物质激发的内向电流可被spantide或CP-99994阻断;GABA激发的外向电流由GABAA和GABAB受体介导;甘氨酸激发的外向电流可被士的宁完全阻断。本研究结果提示DCN神经元既表达兴奋性神经递质受体又表达抑制性神经递质受体,即谷氨酸和P物质介导的外用伤害性信息与GABA和甘氨酸介导的抑制性信息可能在DCN神经元水平进行整合,从而起到了抑制伤害性信息传入的作用。本研究为进一步探索DCN参与伤害性信息的传递和镇痛作用的机理提供了电生理学基础。The responses induced by several putative neurotransmitters involved in nociceptive transmission and inhibition were studied in neurons of dorsal commissural nucleus (DCN) from sacral spinal cord slices. Blind patch-clamp whole-cell recordings were made from 40 DCN neurons. At a holding potential of -60 mV, Glutamate(Glu) and Substance P (SP) evoked inward currents in most of DCN neurons recorded. Gln responses were mediated by both NMDA and non-NMDA sensitive receptors. γ-Aminobutyric acid (GABA) evoked outward currents were mediated by both GABAA and GABAB receptors. Glycine (Gly) induced an outward current which was completely abolished by strychnine. These results suggest that DCN neurons express both excitatory and inhibitory neurotransmitter receptors. The possible function of DCN neurons in visceral and somatic nociception was discussed.
