摘  要:目的:研究芎芷地龙汤对偏头痛模型三叉神经脊束核c-fos和NOS蛋白共存表达的影响。方法:健康雄性SD大鼠随机为4组:生理盐水组、模型组、舒马普坦组、中药组。皮下注射NTG注射剂复制实验性偏头痛动物模型,经药物干预后在4h、6h两个时间点取材,采用免疫组化双标染色法测定三叉神经脊束核细胞癌基因fos(celluar oncogene,c-fos)和一氧化氮合酶(nitric oxide synthase,NOS)的免疫阳性神经元的兴奋状态及其共存表达情况。结果:模型组c-fos阳性细胞数和NOS阳性细胞数增多,4h多于6h,两个时间点的阳性细胞数与生理盐水组比较有显著性差异(P<0.01)。中药组和舒马普坦组c-fos阳性细胞数和NOS阳性细胞数减少,两个时间点的阳性细胞数与模型组比较有显著性差异(P<0.01)。结论:芎芷地龙汤可以减少由硝酸甘油诱导的致痛物质c-fos、NOS的表达,从而进一步减少三叉神经脊束核神经元的兴奋性,阻止痛觉信息的传递。Authors studied the effects of Xiong Zhi Di Long Decoction on the coexistence expression of c-fos and NOS in the spinal nuclei of trigeminal nerves in migraine models. In the study, healthy male SD rats were randomly divided into 4 groups: normal sodium control, model control, Sumatriptan control, and Xiong Zhi Di Long Decoction. Migraine animal models were reproduced through subcutaneous NTG injections. Neuronal emotional state and coexistence expression of c-fos and NOS in the spinal nuclei of trigeminal nerves were determined using immunohistochemical dou-ble staining techniques 4h and 6h after medicinal interference. Modeling results show that positive c-fos and NOS neurons were increased in number in the model control, being significant (P〈0.01)compared with the normal sodium control at 4h and 6h. More neurons were detected at 4h than at 6h. Both Chinese materia medica group and Sumatrip- tan control saw a decreased number of c-fos and NOS neurons, being significant (P〈0.01)compared with the model control at 4h and 6h. It is believed that Xiong Zhi Di Long Decoction is able to reduce the expression of c-fos and NOS induced by NTG. The Decoction can depress neuronal excitability in the spinal nuclei of trigeminal nerves, and inhibit the transmission of nociceptive pain information.
