摘 要:背景与目的端粒酶逆转录酶(hTERT)和p16参与多种肿瘤的发生发展。本研究旨在探讨端粒酶逆转录酶和p16在人非小细胞肺癌组织中的表达情况。方法应用实时荧光定量RT-PCR法和免疫组化SP法检测117例非小细胞肺癌组织及癌旁组织,21例肺良性疾病组织中端粒酶逆转录酶和p16的表达情况。结果hTERT mRNA在非小细胞肺癌组织的表达量为2.937±0.836,明显高于正常肺组织2.042±0.378(t=-5.242,P<0.01);肺良性疾病中p16蛋白表达率为85.7%,高于癌组织中的47.9%,差异有统计学意义(P=0.004)。hTERT mRNA表达与非小细胞肺癌病理类型具有统计学意义(P<0.05),p16蛋白表达与非小细胞肺癌临床分期、淋巴结转移及细胞分化程度具有统计学意义(P<0.05)。hTERT mRNA与p16蛋白表达存在相关性(P<0.05)。结论hTERT mRNA表达对非小细胞肺癌诊断具有潜在临床价值,端粒酶逆转录酶和p16蛋白的表达情况与非小细胞肺癌的发生、发展有关。Background and objective hTERT and p16 are involved in oncogenesis and development of tumor. The aim of this study is to investigate the expression of human telomerase reverse transcriptase (hTERT) and p16 in non-small cell lung cancer (NSCLC). Methods The quantitative reverse transcription-polymerase chain reaction (RT-qPCR) and immuno- histochemistry were applied to detect the hTERT and p 16 in tissue of 21 cases of lung benign diseases and 117 of non-small cell lung cancer and adjacent tissues, respectively. Results hTERT mKNA levels from NSCLC in 117 patients and normal lung tissue in 21 normal controls were 2.937±0.836 and 2.042±0.378, respectively (t=-5.242, P〈0.01). Expression of p 16 protein was observed in 85.7% of normal tissues, while 47.9% of lung cancer tissues showed p16 protein expression (P=0.004). The expression of hTERT mRNA was significantly correlated with the histology (P〈0.0S); the expression of p16 protein was significantly correlated with the clinical stage, degree of differentiation and lymph node metastasis (P〈0.05). The significant correlation between the expression of hTERT and p16 (P〈0.05). Conclusion The hTERT may be useful in clinical diagnosis of NSCLC. Expression of hTERT and p 16 is related to the carcinogenesis and development of NSCLC.
