摘 要:报道一种测定超氧化物歧化酶(SOD)活性的新的极谱分析方法。在0.125 mol/LNH3·H2O-0.75 mol/L NH4 Cl-2.5% NaSO3-0.5%吐温-80(pH= 8.8± 0.3)的介质中,SOD在-0.52V(νsSCE)处产生一氧极谱催化波。SOD在1.0 × 103 U/L~4.0 ×103 U/L的活性含量范围内与催化电流呈线性关系,检出限为8.0×102 U/L。用SOD标准溶液进行标准加入回收测定,平均回收率为99.6%,RSD为0.3%。本方法具有灵敏度高、重现性好、试剂易得、使用安全等特点。A new polyrography for the determination of superoxide dismutase activity was developed. In the medium of 0. 125 mol/L NH3. H2O, 0. 75 mol/L NH4 Cl, 2. 5% Na2SO3 and 0. 5%Tween-80(pH = 8. 8 ± 0. 3), a polarographic catalytic wave of oxygen proceed from superoxide dismutase was observed. The peak potential is - 0. 52 V(vs. SCE). The linear relationship between catalytic current and the activity of superoxide dismutase is from 1 .0 × 103 U/L to 4 .0× 103 U/L. The detection limit is 8 .0× 102 U/L. The recovery is 99.6%. The relative standard deviation (n = 6) is 0. 3%. This method has the advantages of high sensitivity, good reproducibility, and the reagent is accessible.
