摘 要:[目的]确定蒲公英黄酮提取工艺的最佳参数。[方法]以青海省巨大型蒲公英为材料,采用乙醇浸提法提取其黄酮含量,通过单因素试验研究不同乙醇体积分数、料液比(g:ml)、提取温度和提取时间对蒲公英黄酮提取率的影响,再通过正交试验优化提取工艺参数。[结果]单因素试验表明,在乙醇体积分数为60%、料液比为1:25、提取温度为80℃、提取时间为3h时,蒲公英黄酮的提取率最高。正交试验表明,乙醇提取蒲公英总黄酮的影响因素主次为:提取温度>乙醇体积分数>料液比=提取时间,其优化的工艺条件为:提取温度80℃,提取时间3h,乙醇体积分数55%,料液比1:30。[结论]该最佳提取工艺条件经优化验证试验表明,蒲公英的总黄酮得率可达4.89%左右,相对标准偏差3.9%,证明此优化工艺可行。[ Objective ] The purpose was to confirm the optimum parameters for the extraetion process of flavonoids from dandelions. [ Method ] With the giant dandelions in Qinghai Province as iested material, its flavonoids content was extracted by ethanol extracting method, the effects of different ethanol cohen. , the ratio of solid to liquid, the extracting temperature and times on the extraction rate of flavonoids from dandelions were studied through single factor test and the parameters for the extraction process of flavonoids were optimized through the orthogonal test. [ Result ] The single factor test showed that the extraction rate of flavonoids from dandelions was highest when the ethanol concn, was 60%, the ratio of solid to liquid was 1:25, the extracting temperature was 80 ℃ and the extracting times was 2 times. The orthogonal test showed that the factors of affecting the ethanol extraction of flavonoids from dandelions in order were extracting temperature 〉 ethanol conch. 〉 ratio of solid to liquid = extraction time and the optimized process conditions were as follows : extracting temperature of 80 ℃, extracting time of 3 h, ethanol conen, of 55% and the ratio of solid to liquid of 1:30. [ Conclusion] The optimized verification test on the optimum process conditions for the extraction showed that the yield of total flavonoids from dandelions was about 4.89% , with relative standard deviation of 3.9% , proving that the optimized process was feasible.
