摘  要:目的:建立人血浆中依普利酮浓度的HPLC-MS测定法.方法:血浆样品中加入内标溶液后,以液-液萃取方法进行样品处理,采用HPLC-MS方法测定其血药浓度.结果:标准曲线线性范围为2.080~4160μg/L,标准曲线线性关系良好,回归方程为:f=0.0005520+0.001333×C(r=0.9984).高、中、低3浓度的批内和批间变异均小于15%,提取回收率在73.6%~75.5%.结论:建立人血浆中依普利酮浓度的HPLC-MS测定法操作简便,灵敏度高,适用于依普利酮的血药浓度检测及药动学研究.AIM:To develop an HPLC-MS method for the determination of Eplerenone concentration in human plasma. METHODS: The samples were extracted with ethyl acetate after adding the internal standard solution, and analytes were detected by mass spectrometer in positive models. RESULTS: The standard curve was linear in the range of 2. 080 - 4160 μg/L and the mean calibration curve: f= 0. 0005520 + 0. 001333 × C (r = 0.9984). The relative standard derivation of intra- and inter-run was all less than 15%. The extraction recoveries of eplerenone were between 73. 6% - 75. 5%. CONCLUSION: A specific, sensitive and simple method has been established, which is good for determination of eplerenone concentration in human plasma and for pharmacokinetic studies.
