摘  要:目的:探讨2型糖尿病患者红细胞中醛糖还原酶(aldose reductase,AR)的含量与氧化应激指标超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-PX)、丙二醛(malondialdehyde,MDA)水平改变之间的联系。方法:采集25例2型糖尿病患者及25例健康人血样作正常对照,以荧光法检测红细胞中的AR水平,以ELISA法检测血清SOD、GSH-PX活性和MDA含量。结果:2型糖尿病组血清SOD、GSH-PX活性分别为(0.85±0.06)μg/ml和(126.98±1.11)μg/ml,较正常对照组[分别为(12.94±0.11)μg/ml和(298.29±2.56)μg/ml]显著下降(P<0.05),而血清MDA含量则从(1.68±0.05)μg/ml上升至(24.98±2.13)μg/ml(P<0.05),提示2型糖尿病患者的外周血存在氧化应激状态。与此相对应,2型糖尿病患者红细胞中的AR含量为(739.8±80.2)U/gHb,较正常对照组水平[(140.5±26.7)U/gHb]明显升高(P<0.05),其上调表达与氧化应激状态呈正相关。结论:红细胞中AR表达的上调与糖尿病患者体内的氧化应激关系密切,可能通过该环节参与2型糖尿病病变的发生和发展。Objective: To observe the relationship between the content of aldose reductase (AR) in erythrocyte of patients with type 2 diabetes and serum levels of superoxide dismutase (SOD), glutathione peroxidase (GSH-PX) as well as malondialdehyde (MDA), which are biomarkers for oxidative stress response. Methods: The whole bloods were collected from 25 cases of patients with type 2 diabetes. Fluorescence method was applied to detect the AR level in erythrocytes, and ELISA method was performed to analyze the serum levels of SOD, GSH-PX and MDA. 25 blood samples from healthy volunteers were treated in the same way and selected as control. Results: The serum activities of SOD and GSH-PX in diabetes group were (0.85±0.06)μg/ml and (126.98±1.11)μg/ml respectively, showed a remarkable decrease (P〈0.05) compared with those of the healthy control group of (12.94±0.11)μg/ml and (298.29±2.56)μg/ml respectively. While the serum level of MDA increased significantly from (1.68±0.05)μg/ml to (24.98±2.13)μg/ml (P〈0.05), indicating an increasing state of oxidative stress in peripheral blood of patients with type 2 diabetes. At the same time, a remarkable increase of AR level in erythrocytes of (739.8±80.2)U/g Hb of type 2 diabetes was observed, which was significantly higher than that of control group [(140.5±26.7) U/g Hb] (P〈0.05), and the up-regulation of AR expression in erythrocytes had a positive relation with oxidative stress state in peripheral blood. Conclusion: The up-regulated expression of AR in patients with type 2 diabetes is closely associated with oxidative stress state, and may be involved in the occurrence and development of type 2 diabetes through oxidative pathway.
