摘  要:目的观察黄芪甲苷对长波紫外线(UVA)辐射引起的人成纤维细胞增殖活性变化及氧化损伤的影响。方法采用5、10J/cm2的UVA照射培养的成纤维细胞,加入黄芪甲苷干预处理,以四甲基偶氮唑蓝还原法(MTT法)检测细胞活性,比色法检测细胞上清液中的超氧化物歧化酶(SOD)和丙二醛(MDA)含量变化。结果不同剂量UVA照射后24h,成纤维细胞出现增殖活性下降,活性下降程度与辐射强度成正比;黄芪甲苷干预处理可使细胞活性有一定程度恢复。UVA照射组的SOD明显下降、MDA上升,黄芪甲苷处理的细胞上清液中SOD活性明显增加,而MDA下降(P<0.05)。结论黄芪甲苷可在一定程度上抑制UVA引起的成纤维细胞活性下降,其机制可能与其抑制氧化损伤和增强细胞抗氧化能力有关。Objective To investigate the protective effects of astragaloside on the oxidative damage induced by ultraviolet-A (UVA) irradiation and its possible mechanisms. Methods Subconfluent fibroblasts were shammed or irradiated with different dosages of UVA irradiation and treated with astragaloside. Cellular activity was detected by MTT assay. The activity of superoxide dismutase (SOD) and the level of malondialdehyde (MDA) were detected by colorimetric methods. Results The proliferation vality of fibroblasts descended with the U VA irradiation(5,10J/cm^2), and the activities of SOD decreased, the content of MDA was ascensive(P〈0.05). But under the intervention of astragaloside, the above changes induced by UVA irradiation were inhibited (P〈0.05). Conclusion Astragaloside might inhibit the proliferation decreasing which induced by UVA. And its mechanism may have relationship with the ability of strengthening antioxidation and decreasing oxygen radicals.
