摘 要:目的用D141大孔吸附树脂对藏边大黄鞣质进行分离纯化。方法以洗脱参数为主要考察指标,采用UV法测定鞣质的含量。结果原药材经水渗漉,鞣质得率为93.61%,渗漉液用量为药材的15倍量;D141大孔树脂的静态吸附鞣质量为27.5 mg.g-1;适宜洗脱流速为2 BV.h-1;样品上样开始有鞣质渗漏时,大孔树脂吸附量为14.4 mg.g-1;鞣质主要有效部位为10%和30%浓度乙醇洗脱液,洗脱液用量为柱体积的3倍量,总洗脱率为90.07%;单独使用30%乙醇洗脱,干燥后固体物中鞣质的含量为35.5%。结论D141大孔树脂可将藏边大黄鞣质进行有效分离纯化富集。OBJECTIVE To study extraction and purification of tannin in Rheum emodi Wall using D141 Macroporous resin. METHODS Considering elufion parameter to be a main guideline, UV mathod was used to detect the content of tannin in Rheum emodi Wall. RESULTS The seep filtering rates of tannin in Plain medicinal plant were 93.61% ,and the amounts of seep filtering solution was 15 times than that of plain medicinal plant. The static adsorption amount of tannin was 27.5 mg·g ^-1 ;the flow rate was 2 BV· h^- 1 ;adsorption amount of tannin was 14.1 mg·g ^-1 in leakage point;the main effective section of tannin was eluted by 10% and 30% ethanol. Eluent volume was three times of the column volume and total elution rate was 90.07%. The content of tennia was 35.5% using 30% ehtanol as elution. CONCLUSION D141 macroporous resin can be used to purify the tannin in Rheum emodi Wall.
