摘 要:目的研究小归芍超临界提取物对人胃癌BGC-823细胞端粒酶活性和端粒酶逆转录酶基因表达的影响,以阐明其诱导凋亡的可能机制。方法用不同稀释度(0、12.5、25.0、50.0、100.0、200.0mg/kg)的小归芍超临界提取物作用于培养的人胃癌BGC-823细胞,以PCR-ELISA法检测端粒酶活性,western-blotting法半定量检测端粒酶逆转录酶基因的表达。结果小归芍超临界提取物能抑制BGC-823细胞端粒酶的活性和端粒酶逆转录酶基因的表达。结论小归芍超临界提取物诱导人胃癌细胞凋亡的机制可能与其抑制端粒酶的活性和端粒酶逆转录酶基因的表达有关。Objective To study the effects of supercritical fluid extract of Xiaoguishao on the telomerase activity and the expression of human telomerase reverse transeriptase(hTERT) in human gastric cancer cell line BGC-823. Methods BGC-823 cells were treated, with the extract of Xiaoguishao in a concentration of 0, 12.5, 25.0, 50.0, 100.0 and 200.0 mg/kg for 48 h, respectively. The telomerase activity was analyzed by PCR-ELISA assay and the expression of hTERT was detected by western-blotting experiment. Results In the extract of Xiaoguishao treating groups, PCR-ELISA assay showed that the telomerase activity was significantly decreased and the western-blotting assay showed that the expression of hTERT protein was decreased compared with that in control group. Conclusion The extract of Xiaoguishao could suppressed the telomerase activity and inhibited the expression of hTERT in BGC-823 cells. These might be involved in the mechanisms of human gastric carcinoma cells apoptosis induced by the extract.
