摘 要:目的探讨突触素在人食管胃连接部各类环行平滑肌中的分布情况。方法选取河北医科大学第四医院胸外科收治的高位食管中段癌患者8例,在行根治性食管癌切除食管胃吻合术时切除食管胃连接部,分离出食管下括约肌的套索纤维和钩状纤维,以及部分远端食管环行肌和胃底环行肌,应用免疫印迹(western blot)检测突触素在各类环行平滑肌组织中的表达。结果套索纤维、钩状纤维、食管环行肌和胃底环行肌等平滑肌中突触素表达的积分光密度值分别为284.45±73.69,265.39±87.12,248.40±70.80和317.17±96.25,组间差异无统计学意义(F=1.101 8,P>0.05)。结论人食管下括约肌的套索纤维和钩状纤维,以及远端食管环行肌和胃底环行肌中均有突触素表达,但其表达水平无差异。Objective Structure and functional regulation of human esophagogastric junction remains complicated, and abnormality in which may result in many esophageal benign diseases. This study was conducted to analyze the expression of synaptophysin in the human esophagogastric junction quantitatively by western blot. Methods Eight patients undergoing esophagogastrectomy for middle thoracic esophageal carcinoma in the department of thoracic surgery, the Fourth hospital of Hebei medical university were selected, from which smooth muscles of the sling and clasp fibers of lower esophageal sphincter,and the circular layers of the esophagus and gastric fundus were obtained. Then the expression of synaptophysin in these smooth muscles was detected using western blot, respectively. Results Integrated optical density (IOD) value standing for the expression level of synaptophysin in smooth muscles of the sling and clasp fibers, and the circular layers of the esophagus and gastric fundus was 284.45±73.69, 265.39±87.12,248.40±70.80 and 317.17±96.25, respectively. There was no difference among them ( F =1. 101 8, P〉0.05). Conclusion Synaptophysin exists in all of the smooth muscles of the sling fibers and clasp fibers of human LES,and the circular layers of the esophagus and gastric fundus. Expression of synaptophysin in all of them is not different.
