摘 要:目的建立加压蒸制附片中酯型生物碱部位指纹图谱,为附子炮制工艺评价和饮片质量控制提供科学依据。方法采用高效液相色谱法,ZORBAXExtend-C18柱,样品用乙醚-二氯甲烷(1:4)混合溶剂提取,梯度洗脱,检测波长240 nm。结果指定11个共有峰,指纹图谱的精密度、重复性和稳定性均符合指纹图谱要求,采用同一优选工艺方法的加压蒸制附片样品检测的相似度在0.978以上。结论方法可靠,可用于含乌头碱中药及其制品的质量考察。Objective To establish the fingerprint of ester alkaloid fraction in slices of Radix Aconiti Lateralis Preparata prepared by pressurized steaming, and lay a scientific base for the evaluation of processing technique of Radix Aconiti Lateralis Preparata and qualitative control of the slices. Method In the HPLC procedure the chromatography column was ZORBAX Extend-C18 and the samples were extracted with a mixed solvent of aether-dichloromethare ( 1 : 4 ) in gradient elution. The detection wavelength was 240 nm. Result There were 11 general peaks assigned and the accuracy, repeatability and stability of the fingerprint all met the requirement. The similarity of the slice samples detected by the same optimized technique was over 0.978. Conclusion The method is reliable and can be used in the qualitative study of Chinese herbal remedies and the preparations containing aconitine.
