摘 要:目的:探讨盐酸千金藤碱(CH)对急性肝损伤小鼠的保护作用及其机制。方法:建立四氯化碳(CCl4)和D-氨基半乳糖(D-GalN)致急性肝损伤小鼠模型,设CH大、中、小剂量组,阳性对照组(阿拓莫兰),空白对照组和模型对照组。观察CH对小鼠血清转氨酶、一氧化氮(NO)、一氧化氮合酶(NOS)、肝组织氧化指标及肝脏病理学的影响。结果:CH可明显降低CCl4和D-GalN所致肝损伤小鼠的血清转氨酶活性,增强肝脏超氧化物歧化酶(SOD)活性,减少丙二醛(MDA)和NO的含量,降低NOS活性;并缩小肝脏坏死范围。结论:CH对CCl4和D-GalN所致肝损伤有良好的肝保护作用,其机制可能是提高SOD活性、清除自由基,降低NO含量,减少其对细胞和组织的损伤。Objective:To explore the effect of Cepharanthine Hydrochloride (CH) on acute liver injury in mice caused by carbon tetrachloride (CCl4) or D-galactosamine (D-GalN). Methods: Mice were given different dose of CH prior to CCl4 or D-GalN, and ATOMOLAN was chose as positive drug. ALT and AST activities in serum, SOD and NOS activities in liver, MDA and NO contents in liver, and histopathological examination were detected to study the hepatoprotective effect of CH. Results: CH showed significant protection against the hepatotoxic actions of CCl4 and D-GalN, markedly alleviated aspartate aminotransferase (AST), alanine aminotransferase (ALT) level in serum, enhanced superoxide dismutase(SOD), nitric oxide synthase(NOS) activity in liver, reduced Maeic Dialdehyde (MDA), nitric oxide(NO) content. Histochemical investigation showed that CH diminuted necrosis extent and degree in liver tissue, and reduced hydropic degeneration. Conclusion: CH protects against CCl4 and D-GalN induced liver injury in mice, and the mechanism of CH might be enhancing antioxidant enzyme activities, removing free radical and reducing NOS activites.
