摘  要:在pH6.85的0.2mol/LNaH2PO4-Na2HPO4缓冲介质中,盐酸吡格列酮与考马斯亮蓝G250作用,形成离子缔合物,溶液颜色发生明显改变,最大退色波长为570nm,且盐酸吡格列酮的浓度与溶液的退色程度呈良好线性关系,可用光度法测定盐酸吡格列酮。在570nm波长处,盐酸吡格列酮的浓度在1.25×10-6~2.60×10-5mol/L范围内遵守比尔定律,表观摩尔吸光系数ε570为1.26×104L.mol-1.cm-1,检出限为8.20×10-7mol/L。方法具有较高的灵敏度和良好的选择性,用于临床用药中盐酸吡格列酮的测定,结果满意。In a buffer solution of NaH2POa-Na2HPO4 with a pH value of 6.85, pioglitazone hydrochloride reacted with Coomassie brilliant blue G250 to form an ion-association complex. The colour of solution changed obviously, and the maximum fading wavelength was identified at 570 nm, with the fading eolour intensity being proportional to the concentration of pioglitazone hydroehloride. Thereby a sensitive spectrophotometrie method was developed for determination of pioglitazone hydrochloride. The concentration of pioglitazone hydrochloride in the range of 1.25 × 10^-6 - 2.60 × 10^-5mol/L obeyed the Beer's law at a maximum fading wavelength of 570 nm ; the apparent molar absorptivity ε570 was 1.26 × 10^4 L/(mol·cm) ;and the detection limit was 8.20 × 10^-7 mol/L. This method has high sensitivity and fairly good selectivity,and can be applied to the determination of pioglitazone hydrochloride in medicine samples,with satisfactory results.
