酸枣仁汤对小鼠自发活动的影响

摘　　要：目的:研究酸枣仁汤对小鼠自发活动的影响,确定酸枣仁汤的安全有效浓度。方法:在9AM分别连续灌胃给予雄性昆明小鼠60g.kg-1,30g.kg-1,15g.kg-1的酸枣仁汤,2.5mg.kg-1酒石酸唑吡坦溶液和灭菌注射用水7天,用动物行为学视频分析系统记录并分析第1、3、5、7天给药后30min内和9PM30min内自发活动的次数,比较各组小鼠的死亡率,用电子天平称量并比较给药前和10天后的体重,用血球计数仪检测外周血常规并比较红细胞和血红蛋白的计数。结果:各组小鼠生存率均为100%。与空白对照组比较,各给药组在给药前后的体重和停药3天后外周血中红细胞、血红蛋白的数量均无显著性差异,60g.kg-1,30g.kg-1酸枣仁汤组和酒石酸唑吡坦组小鼠在给药第1、3、5、7天9AM给药后30min内自发活动明显减少,差异具有统计学意义(P<0.05,P<0.01),酒石酸唑吡坦组小鼠在第1天9PM检测的自发活动次数明显减少,差异具有统计学意义(P<0.05),60g.kg-1酸枣仁汤组小鼠在停药3天后9AM的自发活动次数明显减少,差异具有统计学意义(P<0.05)。其余各个时间点各给药组小鼠的自发活动次数少于空白对照组,但是差异无统计学意义。结论:60g.kg-1,30g.kg-1的酸枣仁汤可以减少小鼠自发活动次数,并且无明显毒副作用。Objective ：To study influence of suanzaoren decoction on mice＇ s locomotor activity and determine the safe effective concentration of suanzaoren decoction. Methods ： Kunming male mouse underwent continuous intragastrie administration of suanzaoren decoction 60g ·kg^-1 ,30g·kg^-1,15g·kg^-1, Zolpidem Tartrate 2.5mg·kg^-1 and sterile water for injection at 9a. m. for 7 days. Record the mice＇ s locomotor activity frequency within 30 minutes after administration and within 30 minutes at 9p. m. at the 1^st, 3^rd ,5^th and 72 day by animal ethology video analysis system. Compare groups＇mortality, weigh the mice before administration and after 10 days by electronic balance, detect peripheral blood routine to compare the number of RBC and hemoglobin by blood counting instrument. Results ： The survival rate of each group is 100%. Comparing with blank control group, there is no obvious significance between the numbers of RBC and hemoglobin before and after administration in each group. The locomotor aetivities of 60g ·kg^-1,30g·kg^-1 suanzaoren decoction group and Zolpidem Tartrate group decrease obviously within 30 minutes after administration at 9p. m. at the 1^st, 3^rd ,5^th and 7^th day, with signifieant differenee （ P 〈 0.05, P 〈 0.01 ）. There is significantly different （ P 〈 0.05 ） in Zolpidem Tartrate group＇s locomotor activity decrease at 9 p.m. the 1^st day. And There is signifieantly different（P 〈0.05） in 60g ·kg^-1 group＇s locomotor activity decrease at 9 a.m. 3 days after administration. The other locomotor activities decrease are smaller than blank control group, but without significant difference. Conclusion ：60g·kg^-1,30g ·kg^-1 suanzaoren decoction can decrease the number of mice＇ s locomotor activity without obvious toxic and side - effect.
